Abstract
PERFUSION CULTURING METHODS AND USES THEREOF A BSTRACT Provided herein are improved methods of culturing a mammalian cell that include providing conical containing mammalian suspended about % to 5 about 8 0% of the volume of the container of a first liquid culture medium; incuba ting the container for a period of time at about 31 C to about 40 C at a reactor angle of about 5 degrees to about 85 degrees (e.g., about 40 degrees to about 55 degrees) from horizontal and with an agita tion of about 2 0 revoluti ons per minute (RPM) to about 1000 RPM; and continuously or periodically , during the period of time, removing a first volume of the 10 first liquid culture medium that is substantially free of mammalian cells and adding to the first l iquid culture medium a second volume of a second liquid culture medium, wherein the first and second volumes are about equal, and methods of culturing a mammalian cell that include culturing in a gradient perfusion process a mammalian cell suspended in a l iquid culture medium under conditions that generate in the medium a fluid sheer force 15 and dissolved oxygen (O 2 ) concentration that is the same as (or essentially the same as ) that achieved in a gas -permeable coni cal container containing 4% to 4 0% volume of the container of liquid culture medium when p ositioned at a reactor angle of about 5 degrees to 85 degrees (e.g., 40 to 55 from , incubate d at a temperature of about 31 C to about 40 C, and agi tated at a freq uency of 20 about 2 0 revolutions per mi nute (RPM) to about 1000 RPM .
